If I can, I ask them what they want. If I can't ask, I use PDF. It's more universal and prevents someone from editing my work. Also, I like PDF after a fashion, while I loathe Word.
I've done several that required answers be inserted in a particular Excel sheet so that the recipient could create summaries of all responses, do analyses across them, etc. As others have said, if the RFP doesn't say explicitly what is meant, ask.
